Not realy a Pano2Vr problem, but the best are on this forum!

Depends on the kind of ceiling but often there is a darker circle.
Is there a solution for that in PTGui?

On the floor in the corner you can see also an unsharp area.
How to solve that problem?

When shooting a panorama all camera settings should remain the same.

Your camera should be in (M) manual mode.

You meter the scene to find the appropriate shutter speed required for correct "average" exposure.
All things should remain the same for each image in the panoramic scene.

Same Shutter Speed, Aperture Value, ISO, White Balance etc.

Think of the scene as if it was one single shot.

Ton & Smooth: thanks!
Indeed too big contrast between the outside- and inside view.
After correcting that a bit, it's ok now.
